Working of the System
The Truelab™ Real Time quantitative micro PCR system works on four simple processes: Sample Collection, Sample Preparation, Automatic Analysis and Reporting — that can be completed within an hour's time.

Step 1
Sample Collection
Clinical specimen such as whole blood/serum/plasma/other body fluids and sputum in case of TB are collected fresh in the provided tubes. Frozen specimens can also be used.
Step 2
Sample Preparation
PCR requires extraction and purification of nucleic acids using Trueprep™ AUTO Universal Cartridge Based Sample Prep Kit. Completed in under 25 minutes.
Step 3
Automatic Analysis
Purified nucleic acid is loaded onto Truenat™ micro PCR chip and analyzed. Results are generated in less than 30 minutes.
Step 4
Reporting
Results are displayed on screen and printed reports can be generated using the Truelab™ micro printer.
Sample Preparation Process
The PCR process necessitates the extraction and purification of nucleic acids from a clinical specimen, to free it from potential PCR inhibitors. Sample preparation is done using Trueprep™ AUTO Universal Cartridge Based Sample Prep Kit which contains cartridges and reagents necessary for the purpose.
Internal Positive Control (IPC)
The cartridge contains pre-loaded IPC — a full process control that undergoes all the processes the specimen undergoes, from extraction to amplification, thereby validating the analysis from sample to result.

Extraction & Purification Process:
The pre-treated sample is added to the sample chamber of the cartridge. The cartridge is then placed in the Trueprep™ AUTO device for processing.
DNA/RNA from the sample are bound by the matrix enclosed in a cartridge and inhibitors are washed out. At the end, bound DNA/RNA is eluted and collected in elution chamber.
The elute is pipetted using Truepet™ fixed volume precision micropipette into the Elute Collection tube (ECT). Sample preparation completes in under 25 minutes.
Automatic Analysis & Reporting

For the PCR process, test specific Truenat™ Real time quantitative micro PCR chip is loaded onto the chip tray of the TrueLab™ Uno Dx Real Time Micro PCR analyser.
6 µL of the purified nucleic acid obtained from the sample preparation process is pipetted using Truepet™ fixed volume precision micropipette into the Truenat™ Real time micro PCR chip.
At the end of the test, the results are displayed on the screen and printouts can be taken using the Truelab™ micro printer.
Fast Results
The PCR analysis and reporting is completed in less than 30 minutes.

At its core, it uses Polymerase Chain Reaction (PCR) — a highly sensitive molecular biology technique that amplifies specific DNA segments, generating millions of copies from just a tiny amount of starting material.
This enables rapid, accurate detection of pathogens, genetic markers, and other targets with exceptional reliability.
The Three Core PCR Stages
Denaturation
DNA is heated to 94–98°C to break hydrogen bonds, separating the double-stranded DNA into single strands.
Annealing
Temperature is lowered to 50–65°C, allowing specific primers to bind to their complementary target sequences on the single-stranded DNA.
Extension
At ~72°C, DNA polymerase extends the primers by adding nucleotides, synthesizing new complementary DNA strands.
